Abstract
In
Hevea brasiliensis
breeding program, total duration for high yielding clone selection consumed over 25
years. Since molecular marker for identification of latex yield potential in early stage will be beneficial for the
breeder. The main objective of this research was to select the gene marker for identification of latex yielding in
Hevea
brasiliensis
. Gene expression pattern of high and low yielding
Hevea
clone was analyzed using cDNA-AFLP
technique. Up-regulated transcription-derived fragments (TDFs) were selected, cloned and sequenced. The homology
search of the TDFs against the publicly available genes in the database
(GenBank) was performed using the BLAST
sequence alignment program. Eleven candidate genes were obtained; four of them were selected and used to determine
mRNAs levels in latex of four eighteen-months-old high yielding clones and four eighteen-months-old low yielding
clones. The main difference between the high yielding and the low yielding clones was obtained by only TDFLH143
expression pattern. These results indicated that the TDFLH143 gene could possibly be developed as a marker for the
early selection in
Hevea
breeding program.
