The common abnormal Hemoglobins in Thailand when its excluded Hb E and Hb Constant Spring are Hb Hope and Hb Tak. Although, these abnormal Hb cannot lead to severe thalassemia symptoms, Hb analysis of Hb Hope and Hb Tak are complicated interpretation and lead to misdiagnosis from co-migration of Hb Hope and Hb Tak with others Hb. Objective : To develop the Polymerase chain reaction combined with high resolution melting curve analysis (PCR-HRM) for Hb Hope and Hb Tak detection. Methods : Known DNA samples were used for optimization using PCR-HRM. Further analysis of percentage of concordance was done by 97 blind samples for comparison between the developed PCR-HRM assay and allele specific PCR. Results : Our optimization, HRM profile showed specific pattern of melting curve. Further the comparison between PCR-HRM assay and allele specific PCR was done by 97 samples with 100 % accordance. Conclusion : Therefore, our developed HRM should be useful and applied for Hb Hope and Hb Tak detection in thalassemia laboratory.
