Abstract
This research aims to study the denitrification performance in denitrifying bacteria seed culture which
used methanol and fermented fish feed as external carbon source addition at salinity levels of 25 ppm. and to
detect the presence of potential denitrifying bacteria by using Fluorescence
In Situ
Hybridization (FISH)-
technique. It was reported that genus
Methylophaga
spp and
Methylophaga marina
were dominant in the
denitrifying bacterial seed culture. Three durations of semi-batch culture (3, 5 and 7 days) were monitored and
evaluated the denitrification performance and the presence of
Methylophaga
spp. by FISH technique. The results
were found that the longer is the better for denitrification performance (7 days). The qualitative detection of
potential denitrifying bacteria from seed culture by FISH technique were found the presence of Methylophaga spp.
and M. marina. Therefore it was concluded that these bacteria were responsible for the denitrification process in
the denitrifying bacterial seed culture with methanol and fermented fish feed addition.
