ATPS contained 15% PEG and 19.2% Ammonium sulphate salts was the best ratio for cellulose and protease
isolation. Protease was isolated into PEG phase 90.26% and cellulose was isolated into salt phase 78.86%. K
protease
and K
cellulase
were 6.50 and 0.48, respectively. Purified protease and cellulose after partition by using Sepharose 6B
30 cm × 2 cm column, 50 mM Tris-HCl buffer pH 7.0 and flow rate 0.22 ml/min showed that protease had 3.83
purification fold and 48.46 %Yield while cellulase had 2.71 purification fold and 64.04 %Yield. Determined
molecular weight of protease found that there was 2 sub units consist of 20.0 and 19.3 kDa and cellulose had 1 sub
unit contained 7.3 kDa.
