Abstract
The effect of extraction media on the recovery of proteinases from albacore tuna spleen
(
5IVOOVTǰBMBMVOHB
) was investigated. Types of extractants, NaCl concentration and types of surfactants
affected the proteinases extraction. The splenic extract using 50 mM sodium phosphate buffer, pH 7.0
showed higher proteolytic activity than those extracted with distilled water and 50 mM Tris-HCl, pH 7.0
(p<0.05). 50 mM sodium phosphate buffer, pH 7.0 containing different NaCl concentrations were used
to extract the proteinases from albacore tuna spleen. When NaCl concentration was increased (0-1.25
M), the proteolytic activity increased (p<0.05). The splenic extract showed the highest proteolytic
activity when isolated with 1.25 M NaCl. For effect of some surfactants, the highest yield of proteinases
extracted with the aid of 0.2% Brij 35 were obtained. Hence, 50 mM sodium phosphate buffer, pH 7.0
containing 1.25 M NaCl and 0.2% (v/v) Brij 35 was chosen as the extraction medium for proteinases
from albacore tuna spleen.
