Abstract
The purpose of this study was to compare the specific activities of alanine aminopeptidase (AAP) and
dipeptidyl peptidase IV (DPP IV) in saliva of periodontitis patients and periodontally healthy subjects. The
correlation between the enzyme and clinical variables were also evaluated. Ninety Thai subjects were recruited
and divided into 3 groups: group 1) 30 periodontally healthy subjects; group 2) 30 localized chronic periodontitis
patients and group 3) 30 generalized chronic periodontitis patients. All subjects were determined periodontal
disease status by clinical periodontal assessments. Whole stimulated saliva were collected from all subjects for
determining the specific activities of AAP and DPP IV using a colorimetric assay. Median values of the specific
activities of AAP in group 1, 2 and 3 were 0 IU/mg protein. No significant difference was observed among the 3
groups (p=0.57). The correlation between specific activities of AAP and clinical variables were not statistically
significant (p>0.05). Median values of specific activities of DPPP IV in group 1, 2 and 3 were 5.7x10-6, 29.7 x10-6
และ 22.7x10-6IU/mg protein, respectively. The specific activities of DPP IV in group 2 and 3 were significant
higher than group 1 (p=0.001). Moreover, the specific activities of DPP IV was positive correlated with all
clinical parameters (p<0.05). The results showed that periodontitis patients had the higher specific activities of
DPP IV than periodontal healthy subjects. However, the specific activities of AAP among the 3 groups were not
significant difference. These results suggest that the specific activities of DPP IV in saliva might be used as
biomarker for predicting progression of periodontal disease.
